Stabilization of Cysteine-linked Antibody Drug Conjugates with N-Aryl Maleimides.

A research paper was published at Journal of Controlled Release by the joint efforts of SynChem scientists with our partner. The research results show that the newly developed N-aryl maleimide-based antibody-drug conjugate (ADC) will minimize the instability introduced by the retro-Michael addition observed from the regular alkyl maleimide derivatives such as Adcitris and Kadcyla, two marketed ADCs. These data demonstrate that N-aryl maleimide are a convenient and flexible platform to improve the stabilityof ADCs through manipulation of functional group attached to the maleimide ring-head nitrogen.
